Sequence based identification of Verticillium

We suggest the use of elongation factor 1-alpha (EF) gene sequences for species identification. Your sequence can be pasted for species identification here, and PCR conditions you need to generate the sequence are here. If you think you have Verticillium longisporum based on the morphological key and multiplex PCR assay, make sure to read the section below before starting molecular work.

Sequence based identification of Verticillium longisporum

Sequence based identification of Verticillium longisporum is complicated by the fact that V. longisporum is a diploid hybrid and contains two copies of all genes used on VertShield. The PCR primers provided here will thus result in mixed population of PCR amplicons and unreadable DNA sequences. The exception is the ITS region, which has been homogenized due to concerted evolution. However, the ITS region of some V. longisporum isolates is identical to the V. dahliae ITS. For more information on V. longisporum, see Journal paper.
